Những ông bố độc thân kể về một nhóm bạn trai, họ đẹp trai, giỏi giang, nhưng lận đận trong tình duyên. Minh Huy là giám đốc công ty điện thoại. Vợ anh qua đời để lại một cô con gái 17 tuổi đang học tại Nhạc viện thành phố. Tình cảm của cha con Huy gặp nhiều trục trặc.
Anh ghét cay ghét đắng Hương, một đối tác làm ăn rất khó tinh. Hương là bà mẹ đơn thân có một cậu con trai, vốn là bạn thân của con gái Huy. Bạn thân của Huy là Hoàng Quân, giảng viên của nhạc viện. Anh được mệnh danh là một người “ vô tâm nhất thế giới”. Anh có thể đi đón con ở nhà trẻ và rồi để lạc mất con trên đường về. Vợ anh, Nhã Uyên rất thất vọng về anh và đang lung lay vì tình cảm với sếp của mình. Tuy nhiên, Quân lại là một người thầy hết lòng vì học trò.
Hội những ông bố độc thân còn có Vũ Phong - một bác sĩ tâm thần. Anh phải làm “bố” của hai đứa cháu song sinh Cao Nhân và Hiền Đức rất quậy phá. Thành viên trẻ nhất hội là Nguyễn Lâm, chuyên viên điện tử của một cửa hàng bán máy vi tính và laptop. Anh phải một mình nuôi dạy bé Bo mới hơn 5 tuổi. Vợ anh là Hoàng Hoa đang tu nghiệp ở nước ngoài. Tuy là thành viên nhỏ tuổi nhất hội nhưng Nguyễn Lâm lại là ông bố chuẩn nhất.
